Torquetenovirus (TTV), a single-stranded DNA anellovirus, the major component of the human blood virome, shows an increased replication rate with advancing age. An elevated TTV viremia has been associated with an impaired immune function and an increased risk of mortality in the older population. The objective of this study was to analyze the relation between TTV viremia, physical frailty, and cognitive impairment. Methods: TTV viremia was measured in 1,131 nonfrail, 45 physically frail, and 113 cognitively impaired older adults recruited in the MARK-AGE study (overall mean age 64.7 ± 5.9 years), and then the results were checked in two other independent cohorts from Spain and Portugal, including 126 frail, 252 prefrail, and 141 nonfrail individuals (overall mean age: 77.5 ± 8.3 years). Results: TTV viremia ≥4log was associated with physical frailty (OR: 4.69; 95% CI: 2.06–10.67, p < 0.0001) and cognitive impairment (OR: 3.49, 95% CI: 2.14–5.69, p < 0.0001) in the MARK-AGE population. The association between TTV DNA load and frailty status was confirmed in the Spanish cohort, while a slight association with cognitive impairment was observed (OR: 1.33; 95% CI: 1.000–1.773), only in the unadjusted model. No association between TTV load and frailty or cognitive impairment was found in the Portuguese sample, although a negative association between TTV viremia and MMSE score was observed in Spanish and Portuguese females. Conclusions: These findings demonstrate an association between TTV viremia and physical frailty, while the association with cognitive impairment was observed only in the younger population from the MARK-AGE study. Further research is necessary to clarify TTV’s clinical relevance in the onset and progression of frailty and cognitive decline in older individuals.Publication Combined effects of drought and soil fertility on the synthesis of vitamins in green leafy vegetables(2023) Park, Taewan; Fischer, Sahrah; Lambert, Christine; Hilger, Thomas; Jordan, Irmgard; Cadisch, GeorgGreen leafy vegetables, such as Vigna unguiculata, Brassica oleraceae, and Solanum scabrum, are important sources of vitamins A, B1, and C. Although vitamin deficiencies considerably affect human health, not much is known about the effects of changing soil and climate conditions on vegetable vitamin concentrations. The effects of high or low soil fertility and three drought intensities (75%, 50%, and 25% pot capacity) on three plant species were analysed (n = 48 pots) in a greenhouse trial. The fresh yield was reduced in all the vegetables as a result of lower soil fertility during a severe drought. The vitamin concentrations increased with increasing drought stress in some species. Regardless, the total vitamin yields showed a net decrease due to the significant biomass loss. Changes in vitamin concentrations as a result of a degrading environment and increasing climate change events are an important factor to be considered for food composition calculations and nutrient balances, particularly due to the consequences on human health, and should therefore be considered in agricultural trials.Publication Composite flour from Indonesian local food resources to develop cereal/tuber nut/bean-based ready-to-use supplementary foods for prevention and rehabilitation of moderate acute malnutrition in children(2021) Fetriyuna, Fetriyuna; Purwestri, Ratna Chrismiari; Susandy, May; Köhler, Realm; Jati, Ignasius Radix A. The prevalence of undernourishment remains alarming, especially among children under five years old in many countries, including Indonesia. Nowadays, the handling of undernourishment has shifted to treatment outside the hospital, utilizing local nutrient-rich foods. At the national level, the utilization of local food resources is a part of the promotion of dietary diversification and the bioeconomy. Ready-to-use supplementary food (RUSF) refers to supplementary foods aimed at improving the nutrition of moderate acute malnutrition (MAM) children under five years old. RUSF biscuit recipes were made using local food resources available in Banten province, Indonesia. To optimize the nutritional profile of the developed RUSF, taro/talas banten were mixed with ground-nut/peanut (Arachis hypogaea L.) and mungbean (Vigna radiata) as protein and lipid sources and red rice (Oryza longistaminata) and maize (Zea mays) as carbohydrate sources, and enriched by the local banana Nangka (Musa textilia). Two formulations were selected for the pilot testing, namely the taro-peanut and taro-peanut/mungbean RUSF biscuits, made from taro Banten, cereal, peanut and/or mungbean, and local banana. The RUSF biscuit showed promising results, presenting a high level of acceptance and a macronutrient composition that meets the standards for MAM children. However, the RUSF biscuits should be fortified with micronutrient premix to fulfill the dietary requirement for the MAM children. The results of this study provide further development opportunities.Publication Demethoxycurcumin and Bisdemethoxycurcumin are more bioavailable than Curcumin: A meta-analysis of randomized cross-over trials in healthy umans and an In Vitro mechanistic exploration(2023) Desmarchelier, Charles; Sus, Nadine; Marconot, Grégory; Gillet, Guillian; Resseguier, Noémie; Frank, JanBackground: Curcuminoids are secondary plant metabolites found in turmeric and many dietary supplements. They usually consist of a mixture of curcumin (CUR), demethoxycurcumin (dCUR) and bisdemethoxycurcumin (bdCUR). CUR, the main curcuminoid, has been intensely investigated for its putative effects against, e.g., inflammation, oxidative stress and cancer. However, CUR displays very poor bioavailability. We have previously shown that, when brought by turmeric, dCUR and bdCUR, which can also exert health effects, display greater in vitro bioaccessibility than CUR (PMID: 37073511). However, their bioavailability relative to that of CUR has not been thoroughly investigated. Objective: We aimed to compare the bioavailability of dCUR and bdCUR to that of CUR in a meta-analysis of clinical trials in healthy humans and to compare their in vitro bioaccessibility and enterocyte uptake efficiency. Methods and Results: Studies published until 2022 were searched for using Medline and Scopus. The included studies were randomized trials that measured the bioavailability of CUR, dCUR and bdCUR in healthy participants. Estimates were calculated using a random-effects model. Fifteen trials were included in the study, representing a total of 50 interventions, i.e., each trial investigated several curcuminoid formulations, in 762 participants. The relative bioavailabilities were calculated using the inverse variance method. dCUR was 2.32 (95% CI:1.70, 3.13) times more bioavailable than CUR, while bdCUR was 2.57 (95% CI: 1.58, 4.16) times more bioavailable than CUR, with some heterogeneity across the formulations used. Using an in vitro gastro-intestinal digestion model with pure curcuminoids, we showed that dCUR solubilization efficiency was 4.8 and 5.3 times higher than that of CUR and bdCUR, respectively (p < 0.001), while its micellization efficiency was 10.3 and 5.1 times higher than that of CUR and bdCUR, respectively (p < 0.001). Conclusions: bdCUR and dCUR display greater bioavailability in humans compared to CUR. A subgroup analysis by formulation is undergoing investigation and will be presented. For dCUR, this difference is partly explained by higher in vitro bioaccessibility. Uptake efficiency measurements of pure curcuminoids and of curcuminoids from in vitro digestion fluids are undergoing investigation and will be presented. bdCUR and dCUR might therefore represent relevant alternatives to CUR for the systematic delivery of curcuminoids.Publication Evaluation land use cover changes over 29 years in Papua province of Indonesia using remote sensing dataLetsoin, Sri Murniani Angelina; Herak, David; Purwestri, Ratna ChrismiariLand use/cover change (LUCC) observation and determination have been extensively discussed in natural resources management, biodiversity and ecosystem preservation, land management also climate changes studies. An evaluation of the LUCC in Merauke, the easternmost city of Indonesia, was conducted to gain relevant information in agriculture and forestry based on historical data from remotely sensed land cover data. To obtain the historical dynamics of the LUCC, a supervised classification algorithm was implemented to the Landsat images of 1990, 1996, 2009, 2015, and 2019 as well as statistical analysis Wilcoxon signed-rank. This study aims to investigate and analyze the LUCC over a period of 29 years. The results show a significant decline in the forest area by around 7% in 2019. As compared to the LUCC in 1990, agriculture sectors such as dry-land agriculture, shrub-mixed dry land and paddy field gained an area higher than 50%, while the area of the water body is a declined by 0.61 % over the period. The evaluation of LUCC will help government development plans in the protection of commodities in this Province and the promotion for biomass and bioenergy resources.Publication Nutrition during pregnancy and early infancy in urban and rural areas of Deyang region, Southwestern China(2022) Gao, Haoyue; Birner, ReginaIn the past years, China has made remarkable progress in achieving the Millennium Development Goals. However, there are still significant urban-rural disparities and regional differences in the main indicators of child nutrition, maternal and child health. Inadequate dietary intake with regard to certain macro- and micronutrients occurs among women during the reproductive period. Despite the efforts that have been made to promote breastfeeding in China since the 1990s, there is still a very low prevalence of exclusive breastfeeding during the first six months of life. Therefore, the purpose of this observational study was to assess dietary intake and identify risk factors for nutritional inadequacy in pregnant women and to assess the current situation of infant feeding practices during the postpartum hospital stay in urban and rural settings of the study area in China. Between 2010 and 2012, cross-sectional sampling was used in two urban hospitals and five rural clinics randomly selected in the Deyang region. A total of 203 pregnant women in the third trimester and 204 pairs of mothers and newborns were recruited on the basis of informed consent during antenatal clinic sessions and in postnatal wards respectively. Semi-structured interviews on anthropometric and socio-demographic information were conducted for both the pregnant women and postpartum mothers. In addition, data on 24-h dietary recalls of pregnant women, birth outcomes and infant feeding practices of postpartum mothers were collected. Prior to pregnancy, 26.3% of the pregnant women were found to be underweight, while only 5.1% were overweight, based on self-reported pre-pregnancy weight. The women’s overall dietary energy intake during the third trimester originated excessively from fat, was low in carbohydrates, and reached the lower limits for protein. Compared to rural areas, women living in urban areas had significantly higher reference nutrient intake fulfilment levels for energy, fat, protein, vitamin A, Zn, Fe, Ca and riboflavin. The likelihood of pregnant women following certain traditional food recommendations such as avoiding beef, mutton, fish etc. was higher in rural than in urban areas. Both the short-term (e.g. gestational weight gain) and long-term nutritional status (e.g. height) of the mothers affected the birth outcomes (e.g. type of delivery). The decision for a Caesarean Section (CS) was not only related to medical/obstetrical issues but also nutritional and social demographic factors. The short stature of mothers, excessive gestational weight gain, larger-sized newborns, higher educational level or older ages of mothers were significantly positively related to the probability of CS. Although almost all of the mothers started breastfeeding after childbirth, the high prevalence of CS delayed initiation of breastfeeding. Due to the widespread use of infant formula, the prevalence of exclusive breastfeeding (EBF) was only 8.0 % one week after delivery. Regardless of prelacteal feedings, almost exclusive breastfeeding (AEBF) was 34.5%. Breastfeeding initiation within two days after birth was positively associated with AEBF, whereas smaller newborns, mothers with higher education level, mothers lack of knowledge about the importance of colostrum were negatively associated with AEBF. As most mothers lacked knowledge of the lactation physiology, higher formal education was often not related to a better knowledge of breastfeeding. In conclusion, culturally sensitive nutrition education sessions are necessary for both urban and rural women. Special attention must be given to the nutritional status, especially of rural women, before or, at the latest, during pregnancy. Personalized nutritional guidance for expectant mothers should be based on comprehensive consideration of all aspects, such as dietary intake, place of origin (rural vs. urban), body height, and gestational weight gain etc. Better guidance for the gestational weight gain of Chinese mothers needs to be established. Although disparities between urban and rural areas exist, the situation of infant feeding is inadequate in both settings. The high prevalence of CS, the mothers’ poor knowledge of the physiology of breast milk production, the mothers’ lack of breastfeeding confidence, the widespread advertising of breast milk substitutes, and the changing perception of the function of breasts, may influence the unfavorable breastfeeding behavior observed in the study area. In order to prevent CS and to promote breastfeeding, education about the complications and disadvantages of CS especially among mothers with higher education, older childbearing age, and primiparas needs to be strengthened. Supporting adequate maternal nutrition during the reproductive period and creating a good breastfeeding environment requires the efforts of the whole society.Publication Tocochromanol profiles in Chlorella sorokiniana, Nannochloropsis limnetica and Tetraselmis suecica confirm the presence of 11′-α-tocomonoenol in cultured microalgae independently of species and origin(2022) Montoya-Arroyo, Alexander; Lehnert, Katja; Muñoz-González, Alejandra; Schmid-Staiger, Ulrike; Vetter, Walter; Frank, Jan11′-α-Tocomonoenol (11′-αT1) is structurally related to vitamin E and has been quantified in the microalgae Tetraselmis sp. and Nannochloropsis oceanica. However, it is not known whether 11′-αT1 is present in other microalgae independent of species and origin. The aim of this study was to analyze the tocochromanol profiles of Chlorella sorokiniana, Nannochloropsis limnetica, and Tetraselmis suecica and to determine if 11′-αT1 is present in these microalgae. Cultured microalgae were freeze-dried and the presence and identity of α-tocomonoenols were confirmed by LC-MSn (liquid chromatography coupled to mass spectroscopy) and GC-MS (gas chromatography coupled to mass spectroscopy). Tocochromanol profiles were determined by HPLC-FLD (liquid chromatography with fluorescence detection) and fatty acid profiles (as fatty acid methyl esters; FAME) by GC-MS. As confirmed by LC-MSn and GC-MS, 11′-αT1 was the dominant αT1 isomer in cultured microalgae instead of 12′-αT1, the isomer also known as marine-derived tocopherol. αT1 represented less than 1% of total tocochromanols in all analyzed samples and tended to be more abundant in microalgae with higher proportions of polyunsaturated fatty acids. In conclusion, our findings confirm that αT1 is not restricted to terrestrial photosynthetic organisms, but can also accumulate in microalgae of different species, with 11′-αT1—and not the marine-derived tocopherol (12′-αT1)—as the predominant αT1 isomer.
